Even though Vayne is not typical support, you are gonna support your team very effectively. With heals, shields and buffs:
Heal summoner spell.
Guardian and Font of Life runes.
Redemption, Locket of the Iron Solari and Moonstone Renewer and other items.
Playstyle
🛡️ Play safe, stay in the back and play like support, so you can stay and heal more.
⚔️ Sometimes, you can go in and play more aggressively when the opportunity is right.
💖 Focus on healing and shielding to keep allies alive.
✨ Use Redemption, Locket, and Heal on cooldown for healing.
How to play
General
Your first rush is Redemption.
Your second rush can be Locket of the Iron Solari (vs AoE), Moonstone Renewer (sustain).
Also, Echoes of Helia is pretty good because you can proc it consistently with Font of Life (slow/cc).
Rush second Locket if your team already doing teamfights or when an enemy team has champions with super aoe damage (example: like Fiddle's ult). Otherwise, get Moonstone Renewer #2. The third item should be whatever you did not buy - Locket of the Iron Solari or Moonstone Renewer. That's your core.
Another decent item if you have carry/strong bruiser/strong tank is Knight's Vow.
Try to use summoner spell Heal on cooldown. With Moonstone Renewer, ideally there should be at least two people for maximum healing effect.
Later in the game, for maximum support - you can sell boots for sixth item.
You can get items like Ardent Censer (some auto attackers needed) or Staff of Flowing Water. It will be triggered often enough that it will be worth.
Interesting item is Cryptbloom. If you want to go full healing, you can get it like #4+ item.
Runes
With Cosmic Insight rune from Inspiration tree:
Redemption and Locked of the Iron Solari has 81.92 sec cooldown (down from 90 sec).
Shurelya's Battlesong has 68.12 sec cooldown (down from 75 sec).
Guardian is important rune. It is going to provide tons of shielding. Remember that you need to be close to your allies to proc it.
You can also manipulate it - for example, you can stick to one ally, or avoid some spells that's gonna only waste it.
You will be triggering Font of Life effect, which will heal your allies. It also chains off Moonstone Renewer!
Font of Life total healing is not properly calculated in a post-game lobby. Value is bigger in the game vs. what's displayed in statistics. You heal more!
Another important rule is Cosmic Insight, it reduces cooldown for your active items (so more heals/shielding) and Heal summoner spell.
Redemption
Try to use Redemption on cooldown, when your allies are missing some health. It is best to use when they are grouped. Especially if you already have Moonstone Renewer, it is best to wait (or ping) until they group together, so you can have them all healed.
Remember that you can use Redemption for the damage. However, in general, it is not worth it. Keep it to heal your team!
Be careful when using Redemption while you are dead, and you are about to be respawned. The camera will go to your position, and you can cast it in the spawning pool by mistake. Watch out for bad timing!
Disrupting
You can sow chaos in the fights, pretending that you are going all in on enemy carry, going invisible, condemning enemies into walls, getting focus and disrupting fights. And then getting out with Q.
Enemies won’t expect Vayne support. They will assume you’re ADC, so they will focus you in fights - use that to bait cooldowns or redirect pressure from your actual carries.
Peeling
You can be really great peeler. If you peel, you are often not actively focused, meaning free auto hits and serious Silver Bolts (W) damage. You can Condemn (E) into walls or knock enemy away or even further into your team.
Damage
Even though you are not building any damage items, you will always be a threat thanks to your kit thanks to Silver Bolts (W).
Condemn (E)
Your Condemn (E) can be used defensively to peel for your carry or offensively to punish overextending enemy. Hitting a stun on the wall can turn fights instantly.
Tips and Tricks
Skill Order
Generally speaking, Vayne skill order is flexible. When you need more mobility and manoeuvrability, max Tumble (Q). When you are dealing with tanks and bruisers, max Silver Bolts (W).
Tumble (Q)
Don't be predictable with your Tumble (Q) and don't tumble forward all the time! Tumble backward, diagonal, sideways. This way, your enemies will have hard time guessing your position.
You can have instant Tumble (Q) if you position yourself next to terrain, wall or turret. This way you can push faster, proc Silver Bolts (W) faster or win duels (as you auto attack faster this way).
Condemn (E)
Walls, obstacles and terrains created by champions (both allies and enemies) counts for your Condemn (E)!
Final Hour (R)
You can hide Final Hour (R) animation by casting it in the bush or in fog of war. This way, enemies do not know that you ulted and you have advantage of first strike.
You can elevate your Vayne game immediately just by changing direction with your Final Hour (R). Pretend that you are going one direction, just to R + Q into another.
Engaging
You can start fights for your team. Combo of Final Hour (R) + Tumble (Q) + Condemn (E) (hide the animation!) enables you to do excellent engages and catch people off. Then safely Q away with invisibility.
To do it, you can R + Q from bushes or fog of war, then pin enemy into the wall for an easy kill.
Riot
Tumble has many uses, but it cannot pass over walls.
Condemn can be used to both pin targets to walls to ensure a kill, or to escape a pursuer.
Don't go in first to a large teamfight. Wait for your teammates to initiate.
